Introduction: EDTA is one of the most commonly used irrigators in endodontic practice, however, its chelating action causes damage to dentin structure. Apple vinegar has been regarded as a substitute for EDTA. Because of that, the present study aimed to evaluate the apple vinegar action on dentin surface, of human and bovine teeth, used alone or in association. Materials and Methods: Forty-eight human and 48 bovine teeth were divided into twelve groups irrigated for 3 min with 10 ml of a solution (Saline: Saline solution; NaOCl: 2% Sodium hypochlorite; AV: apple vinegar; EDTA: 17% EDTA; NaOCl + AV: 2% sodium hypochlorite + apple vinegar; NaOCl + EDTA: 2% sodium hypochlorite +17% EDTA). After irrigation teeth were sectioned longitudinally and evaluated in SEM to observe smear layer removal; tubule area and inter and peritubular erosion. The measurement area was performed in 5000x magnification using ImageJ software for erosion analysis and the presence of smear layer images were used at 7500x and 2000x and a scoring system used for evaluation. Data were subjected to Kruskal-Wallis and Dunn post-hoc. Tubule area measurements were subjected to one-way ANOVA and Tukey post-hoc at α= 0. 05.Results: Smear layer removal was worst in the apical third of human teeth regardless of the substance. AV, when used alone, showed no statistical differences in relation to EDTA. However, when used in combination with NaOCl, it showed a decrease on smear layer removal. The NaOCl and Saline groups showed lower erosion at both inter and peritubular dentin. Dentinal tubules were greater at the cervical than the apical third, being the major difference at NaOCl + EDTA group when human teeth were used, whereas AV group performed better with bovine teeth. Conclusion: Apple cider vinegar can be a substitute of EDTA when larger volumes are used. Bovine teeth can be used for in vitro studies, with similar results to humans.
